Immunohistochemistry analysis of expression of PTEN, p53 suppressor gene in hepatocellular carcinoma

摘要: 为探讨肝细胞肝癌组织中抑癌基因PTEN及 p5 3蛋白的表达情况及临床病理意义。应用免疫组织化学技术检测了 41例肝细胞肝癌及其相应的癌旁组织中PTEN和p5 3蛋白的表达情况。 41例癌旁组织PTEN全部阳性表达 ,肝细胞肝癌组织中PTEN阳性表达率 39% ,阳性信号显示于胞浆中。p5 3阳性表达率 5 1% ,PTEN蛋白在肝细胞肝癌组织中的阳性表达与组织分化程度明显相关 ,高分化癌的阳性率为 73 % ,低分化癌阳性率 2 7%。肝细胞肝癌组织中存在较高比例的PTEN蛋白阴性表达 ,说明在肝细胞肝癌的发生发展中PTEN基因失活起着重要作用 ,它的阳性表达可能有一定的预后意义Abstract: To investigate the expression of the protein of tumor suppressor gene PTEN, p53 in hepatocellular and its clinicopathology.Paraffin embedded tissues from hepatocellular carcinoma 41case and their adjoining tissue were analysed for PTEN, p53 protein expression by immunohistochemical method.Adjoining tissues of 41 carcinoma were all positive for PTEN protein.In the 41 hepatocellular carcinoma, 39% were positive for PTEN protein.The positive signals were seen in the cytoplasm 51% were positive for p53 protein.There was a significant difference between the pattern of expression of PTEN protein in well differentiated carcinoma (73%PTEN positive) and poorly differentiated carcinoma (27% PTEN positive) .There was a higher incidence of negative expression of PTEN protein in hepatocellular carcinoma, which indicates that PTEN gene inactivation may play an important role in the genesis and development of some hepatocellular carcinoma.PTEN protein positive may be feature for favorable prognosis in the patient with hepatocellular carcinoma.
