Stromelysin-1 proenzyme expression in the processe of experimental liver fibrosis.

摘要: 研究实验性肝纤维化过程中基质分解素 - 1(MMP - 3)酶原蛋白表达的动态变化。建立四氯化碳中毒性大鼠肝纤维化模型 ,采用半定量Western免疫印迹方法 ,对肝纤维化不同阶段肝内MMP - 3的酶原蛋白的表达水平进行了检测。在正常大鼠肝组织中有MMP - 3的酶原表达。在肝纤维化发生发展过程中MMP - 3酶原蛋白表达水平逐渐增强 ,至肝纤维化中期到达最高峰 ,到肝纤维化晚期及肝硬化阶段其表达较中期下降 ,但仍高于正常对照水平。MMP - 3在肝纤维化的发生、发展直至肝硬化形成过程中可能发挥着重要作用

Abstract: To investigate the dynamic alteration of stromelysin-1 (metalloproteinase-3, MMP-3) proenzyme protein expression in the processes of experimental liver fibrosis. The carbon tetrachloride-induced hepatic fibrosis rat models were used. MMP-3 proenzyme protein expression in the different stages of liver fibrosis was measured by half quantitative analysis of Western blot. MMP-3 proenzyme protein expression could be detected in normal rat liver. In the pathogenesis and development of liver fibrosis, MMP-3 proenzyme protein expression level increased gradually, with the expression rate being maximum at an intermediate stage of fibrosis. And it decreased gradually during the late liver fibrosis and at thestage of liver cirrhosis, but remained relatively high levels above normal. MMP-3 might participate in pathogenesis of liver fibrosis and cirrhosis as an important role.-
