Expression of RTN gene and its role in oncogenesis of human hepatocellular carcinoma.

摘要: 为研究肝癌中内质网蛋白基因 (RTN)表达及初步功能。利用免疫组化、真核转染、体内外抑瘤试验等方法。结果显示肝癌中RTN呈下调表达 ,RTN蛋白主要定位于细胞浆和细胞膜 ,并且与肝癌的淋巴结转移有关。转染RTN后 ,体外肝癌细胞生长率明显减少 ,一些基因表达发生改变 ,体内肿瘤抑制率为 85 6 4 %。转染RTN细胞产生肿瘤的坏死级别较高于其它两组。RTN可能作为候选抑癌基因在肝癌发生中起重要作用

Abstract: To investigate the expression and role of RTN in the oncogenessis of human hepatocellular carcinoma (HCC) .The expression of RTN in HCC were observed by immunohistochemical staining, eukaryotic transfection tumorigenicity. Among samples of HCC, RTN were down-regulation expression. RTN protein was mainly localized in cytoplasm and cell membrane. The position rate of RTN were 20%?100% in lymph node metastasis and no lymph node metastasis, respectively. The growth rate of the tumor cell were decreased. Some gene expression was changed before and after transfection of RTN. Tumor inhibiting rate was 85.64% through RTN. Sections of tumors were observed, the degree of tissue necrosis by transfection RTN was higher than other groups. RTN could be a candidate tumor suppressor gene for HCC.-
