Comparative study of the effect of two different herbal compounds on CCl4 induced liver fibrosis in rats.

摘要: 比较复方 86 1与复方 96 1对四氯化碳实验性肝纤维化的防治效果。Wistar大鼠随机分为 4组 :对照组(10只 )、四氯化碳模型组 (2 0只 )、复方 86 1组 (15只 )、复方 96 1组 (15只 )。实验 8周 ,取肝组织 ,应用半定量逆转录多聚酶链反应方法 (RT -PCR)检测肝组织Ⅰ型前胶原、Ⅲ型前胶原、TGF - β1、TIMP - 1的mRNA水平。病理组织染色 ,记分。复方 86 1、复方 96 1显著降低Ⅰ、Ⅲ型前胶原、TGFβ1和TIMP - 1mRNA水平 (P <0 0 1)。肝组织病理纤维化分级表明 :两复方抑制纤维结缔组织增生程度相似。复方 96 1与复方 86 1抗纤维化效果相近

Abstract: To compare the anti-fibrotic effects of two Recipes of commonly used Chinese traditional herbal compounds.Sixty male Wistar rats were divided into four groups:control group (10 rats) , model group (20 rats) , group treated with compound 861 (15 rats) and group treated with compound 961 (15 rats) .Hepatic fibrosis was induced by carbon tetrachloride (CCl4) subcutaneous injection for 8 weeks.Herbal compounds 861 and 961 were administered by gastric gavage respectively.At the end of 8 weeks, the gene mRNA level of procollagen Ⅰ, procollagen Ⅲ and transforming growth factor beta one (TGF-β1) and tissue inhibitor of metalloproteinase one (TIMP-1) was examined by means of reverse transcription polymerase chain reaction (RT-PCR) .Histopathology of each group was assessed with HE, Masson and Sirus red staining.Compound 861 and 961 could decrease all mRNA level: procollagen Ⅰ, procollagen Ⅲ mRNA level, TGF-β1 and TIMP-mRNA level (P<0.01) .Compound 861 and Compound 961 could significantly suppress the fibrotic proliferation by pathological assays.It concluded that Compound 861 has the similar antifibotic effect with Compocmd 961. -
