Study of the Survivin expression in HepG2 Cells induced by mitomycin

摘要: 研究肝癌细胞HepG2内凋亡抑制基因Survivin在化疗药物丝裂霉素作用下的表达变化,探讨肝癌细胞可能的耐药机制。在体外培养的肝癌细胞株HepG2中加入低浓度丝裂霉素,于不同时间收集细胞后运用RT-PCR以及免疫印迹法检测Survivin的表达变化。发现加入丝裂霉素12小时未见Survivin明显表达改变,24、48小时后SurvivinmRNA表达较加药前分别增加1.33、1.93倍,蛋白质表达增加1.25、2、10倍。丝裂霉素诱导肝癌细胞Survivin表达升高具有时间效应,Survivin表达升高可能在肝癌细胞耐药过程中发挥了一定作用。

Abstract: To study the Survivin expression in HepG2 cells induced by mitomycin and explore the possible mechanism of drug resistance of hepatocellular carcinoma.Low concentration mitomycin was added to cultured HepG2 cells.Survivin expression was detected by RT-PCR and Western-blot.Surviving expression had no change with addition of mitomycin in 12 hour.Its mRNA expression reached 1.33 times and 1.93 times and protein expression reached 1.25 times and 2.10 times in 24 hour and 48 hour after drug addition.Raised expression of survivin gene in hepatocellular carcinoma induced by mitomycin had time effectors.Rasised expression of Survivin gene had possible effect in the drug resistance of hepatocellular carcinoma.-
