摘要

摘要: 目的探讨聚集素（Clusterin）蛋白在肝细胞癌（HCC）中的表达及其与P糖蛋白（P-glycoprotein,P-gp）表达的相关性。方法采用免疫组化S-P法检测凋亡抑制基因Clusterin在HCC、肝硬化和正常肝组织中的表达及P-gp蛋白在HCC中的表达。结果Clusterin蛋白在HCC中的阳性表达率为82.93%,在肝硬化和正常肝组织中表达阴性或弱阳性;Clusterin蛋白阳性表达与患者的年龄、性别及肝硬化无关,与Edmenson组织学分级有相关性。P-gp蛋白在肝癌组织中的阳性表达率为70.73%。在HCC中,Clusterin蛋白的表达与P-gp蛋白表达呈正相关。结论Clusterin蛋白在HCC中呈高表达,与临床耐药密切相关,有望成为肝癌靶向治疗的一个新靶点。

Abstract: Objective To study the expression of clusterin protein and its relationship with expression of P-glycoprotein in human hepatocellular carcinoma (HCC) .Methods The expression of clusterin gene in HCC, cirrhosis and normal tissue and the expression of P-glycoprotein in HCC were detected by S-P immunohistochenical method.Results The positive expression rate of clusterin protein was 82.93% in HCC, and negative or weak in cirrhosis and normal liver tissue.The positive expression rate of clusterin protein was significantly correlated with Edmondson's histological grade, but was no correlated with patients'age, sex and with or without cirrhosis.The positive expression rate of P-glycoprotein in HCC were 70.73%.There was a significant correlation between the expression of clusterin protein and P-glycoprotein.Conclusion The high expression rate of clusterin is significantly associated with clinical multidrug resistance in HCC.It could be a potential target for treatment of HCC.
