The Changes in substance P in jejunum of fatty liver

摘要: 目的建立脂肪肝大鼠模型,观察脂肪肝胃肠动力紊乱模型中大鼠空肠组织内P物质(SP)的分布及表达,以探讨SP在脂肪肝胃肠功能紊乱发病机制中的可能作用。方法SD大鼠30只,随机分为正常对照组(n=10)和脂肪肝组(n=20)。采用高脂饮食(主要成分为胆固醇、猪油)制备大鼠脂肪肝模型,复制过程中病理组织切片观察组织成模情况,3个月后采用葡聚糖蓝-2000灌胃,作为胃肠内标志物,观察大鼠肠道传输速度,取正常对照组及模型组大鼠空肠标本,应用免疫组化染色法对SP免疫反应阳性神经纤维进行染色,并应用彩色病理图像分析软件进行分析。结果与正常大鼠相比,脂肪肝大鼠小肠动力明显减弱(P<0.05),空肠SP免疫反应阳性纤维表达减弱(P<0.05)。结论脂肪肝大鼠模型中肠运动功能减退与P物质的减少有一定的相关性。Abstract: Objective To observe the expression of substance P (SP) in the mesenteric plexus of jejunum in a fatty liver rat model, therefore, investigate their possible role in the pathogenesis of gastrointestinal dysfunction in fatty liver.Methods Thirty SD rats were selected and randomly divided into the normal control group (n=10) and the fatty liver model group (n=20) .Fatty liver model were fed with high-fat diet.The hepatic tissue was sliced for pathological observation.The blue dextran-2000 was used to label gastrointestinal tract for gastric lavage to quantify the enteric transmission rate after three months.The tissue specimens were obtained from the control and the fatty liver model groups.The expression of SP-positive fibers was analyzed by immuonhistochemistry.The results were examined by means of color image analyzer.Results Compared with that of the controls, the small intestinal transmission rate of fatty liver rats was significantly reduced (P < 0.05) , the expressions of SP decreased (P < 0.05) .Conclusion The decrease of substance P in the mesenteric plexus of jejunum by the fatty liver rat model has a relationship with the decrease of movement of intestine.
