Effects of Ruangan Huajian granule on hepatic expression of α-SMA, TGFβ1, and COL-Ⅰ in C57 mice with experimental hepatic fibrosis

目的观察软肝化坚颗粒对肝纤维化模型C57小鼠肝组织中α-平滑肌肌动蛋白(α-SMA)、转化生长因子(TGF)β1表达和I型胶原水平的影响。方法 40只C57小鼠随机分为4组,分别为正常对照组、模型对照组、软肝化坚颗粒低剂量组及高剂量组,每组10只。采用四氯化碳(CCl4)腹腔注射制造小鼠肝纤维化模型,通过免疫组织化学染色方法检测肝组织中α-SMA、TGFβ1和I型胶原的表达,同时进行肝组织纤维化评分(S)。结果各模型组肝组织α-SMA、TGFβ1及Ⅰ型胶原表达和纤维化评分与正常对照组比较,差异均具有统计学意义(P<0.05);软肝化坚颗粒低剂量组、高剂量组肝组织α-SMA和TGFβ1及Ⅰ型胶原评分和纤维化评分较模型对照组均明显降低,差异具有统计学意义(P<0.05);软肝化坚颗粒低剂量组和高剂量组α-sma、tgfβ1及ⅰ型胶原评分及肝纤维化评分之间差异无统计学意义(p>0.05)。结论软肝化坚颗粒可能通过抑制C57小鼠肝组织TGFβ1的产生,进而抑制肝星状细胞(HSC)的活化,减少细胞外基质(ECM)的合成,从而降低I型胶原增生,达到预防和治疗肝纤维化的作用。
Abstract:Objective To observe the effect of Ruangan Huajian granule on hepatic expression of alpha-smooth muscle actin (α-SMA) , tumor growth factor-beta 1 (TGFβ1) , and collagen I (COL-Ⅰ) using a C57 mouse model of experimental hepatic fibrosis.Methods Forty C57 mice were randomly divided into four groups (n=10 each) : uninduced normal control group, model control group, model treatment group with low-dose (0.1 g/day for six weeks) Ruangan Huajian granule, model treatment group with high-dose (0.2 g/day for six weeks) Ruangan Huajian granule.Hepatic fibrosis was induced by intraperitoneal injection of 10% carbon tetrachloride.The expression of α-SMA, TGFβ1, and COL-Ⅰ in liver tissue of mice was observed by immunohistochemistry.The liver tissues were also scored for fibrosis stage by evaluating morphological changes detected upon hematoxylin-eosin and Masson trichrome staining.Results There were significantly higher levels of α-SMA, TGFβ1, and COL-Ⅰ expression and scores of fibrosis stage in the hepatic fibrosis mice than in the normal control mice (P<0.05) and in the model control group than in either the low-dose or high-dose model treatment groups (P<0.05) .there="" was="" no="" significant="" difference="" between="" the="" low-dose="" model="" treatment="" group="" and="" high-dose="" p="">0.05) .Conclusion Ruangan Huajian granule may alleviate the degree of fibrosis in C57 mice with experimental hepatic fibrosis by reducing hepatic expression of α-SMA and TGFβ1.Furthermore, Ruangan Huajian granule may depress hyperplasia by reducing COL-Ⅰ expression.
