Role of liver sinusoidal endothelial cells in the pathogenesis of nonalcoholic fatty liver disease

    随着非酒精性脂肪性肝病(NAFLD)在西方发达国家和我国富裕地区的发病率逐年上升,与其相关的治疗手段也在不`断地提高,日益成为人们关注的研究热点。而肝窦内皮细胞(LSEC)作为肝脏的重要生理结构,在NAFLD的发生和发展中发挥着重要的作用。总结了LSEC在NAFLD发病过程中的作用机制,主要包括以下途径:LSEC毛细血管化在单纯性脂肪肝中发生并促进脂肪变性;同时,LSEC有助于非酒精性脂肪性肝炎的氧化应激,并且是非酒精性脂肪性肝炎中肝脏炎症的主要效应物,因此也促进肝纤维化;另外,在NAFLD中,血管生成受到高度刺激并促进NAFLD相关的肝细胞癌发生。然而,LSEC在非酒精性脂肪性肝炎相关性肝硬化中的作用尚未得到具体验证,需要进一步明确,以期为治疗提供新的思路和方向。
