The expression of apoptosis by TNF-α, TNFR and Bcl-2 family in chronic liver disease

摘要: 探讨凋亡蛋白TNF -α、TNFR及Bcl- 2家族在慢性肝炎、肝硬化及肝癌发生、发展中对凋亡的调节作用。 94例慢性肝病患者 ,慢性肝炎 2 8例 ,肝硬化 2 2例 ,肝癌 4 4例 ,采用原位杂交法检测TNF -α、TNFR及免疫组化SP法检测Bcl- 2家族凋亡蛋白及Fas、FasL在其肝组织中的表达。肝硬化TNF -α、TNFR表达率为 90 9%、95 5 % ,明显高于慢性肝炎组 (5 3 6 %、4 6 4 % )及肝癌组 (43 2 %、38 6 % ) ,P <0 0 5或 <0 0 1;Fas在慢性肝炎和肝硬化与肝癌组表达分别为 (10 0 0 %、81 8%、5 6 8% ) ;前二者与肝癌相比P <0 0 1表达明显高于肝癌组。HAI记分越高TNF-α、TNFR和Fas表达越明显 ,而Bcl- 2表达越低。说明在慢性肝病时 ,TNF -α、TNFR、Bcl- 2各凋亡蛋白家族参与了肝组织凋亡的调节 ,引起不同程度的肝组织损伤Abstract: To study the expression of TNF-α?TNFR and Bcl-2 family in patients with chronic hepatitis?liver cirrhosis and hepatocellular carcinoma. The apoptosis family in the liver tissue was studied by Nucleic acid molecular hybrodisation or immunohistochemistry. In 94 cases of TNFR in the liver cirrhosis group were significantly higher than that in chronic hepatitis and hepatocellular carcinoma group ( P <0 01) . The greater of expression level of TNF-α?TNFR and Fas in higher HAI. On the contrary, The lower of expression level of Bcl-2. It is suggested that TNF-α?TNFR and Bcl-2 famlily is involved in the regulation of apoptosis in the chronic liver diseases.
