The realationship among activation-induced cell apoptosis of peripheral blood lymphocytes with the level of HBVDNA in the serum and Th1/Th2-type cytokins production in chronic hepatitis B virus infection patients

摘要: 研究活化诱导的外周血单核细胞凋亡 （AICD）在乙型肝炎慢性化中的作用以及与HBVDNA和Th1/Th2的关系。分离 2 5例慢性乙型肝炎患者与 10例健康人外周血单个核细胞 （PBMC） ,在PHA的刺激下培养 ,经流式细胞仪检测凋亡 ,ELISA检测细胞因子IFN -γ、IL - 4、IL - 10。经过PHA诱导的PBMC凋亡率乙型肝炎组明显高于正常对照组 （P <0 0 1）。在慢乙肝组中 ,随着HBVDNA的上升 ,凋亡比率升高 ,IL - 4与凋亡成正相关 ,有统计学差异（P <0 0 0 1）。AICD可能在乙型肝炎感染慢性化与形成免疫耐受过程中起到重要作用 ,HBVDNA与AICD有关 ,IL -4促进AICD的发生Abstract: To research the realationship among activation-induced cell apoptosis of peripheral blood lymphocytes with the level of HBVDNA in the serum and Th1/Th2-type cytokins production in chronic hepatitis B virus infection patients. PBMC of 25 chronic hepatitis B virus infection patients and 10 healthy people are separated and cultivated with PHA. The activation-induced cell apoptosis of the patients with hepatitis B was measured with flow cytometer. the productions of IFN-γ、IL-4、IL-10 were examined by ELISA. Compared with the controls, activation-induced cell apoptosis is increased significantly in chronic hepatitis B virus infection patients, with the development of the level of HBVDNA, the level of activation-induced cell apoptosis is increased obviously, the production of IL-4 has a positive effect to AICD. AICD plays an important role in the persistence of chronic hepatitis B virus infection patients, The level of HBVDNA and IL-4 have a positive effect on activation-induced cell apoptosis of peripheral blood mononuclear cells.
