Study on pathological significance in the expression of the nerve growth factor (NCF) and its mRNA in benign and malignant lesions of gallbladder.

摘要: 目的研究胆囊良恶性病变组织中NGF及其mRNA表达水平和临床病理意义。方法108例胆囊腺癌、46例癌旁组织和35例慢性胆囊炎标本常规石蜡包埋切片,NGF染色方法为En VisiomTM免疫组化法,NGFmR-NA染色方法为原位杂交法。结果胆囊腺癌NGF和NGFmRNA表达阳性率及其评分明显高于癌旁组织和慢性胆囊炎组织(P<0.01);腺瘤癌变或高分化腺癌、肿块最大径<2cm、无淋巴结转移和未侵犯周围组织病例NGF和NGFmRNA表达阳性率及其评分明显低于低分化腺癌、肿块最大径≥2cm、淋巴结转移及侵犯周围组织病例(P<0.05或P<0.01)。结论NGF及其mRNA表达水平可能与胆囊腺癌发生、进展、侵袭有密切关系,可能是反映胆囊癌预后的重要生物学标记物。Abstract: Objective To study the expressive levels and clinicopathological significances of NGF and NGF mRNA in the benign and malignant lesions of gallbladder.Methods EnVisionTM immunohistochemical method for detection of NGF and in situ hybridization for expressive levels of NGF mRNA was used in the routinely paraffinembedded sections from patients of gallbladder adenocarcinoma (n-108) , paraneoplastic tissues (n=45) and chronic cholecystitis (n=35) .Results The positive rates and scores of NGF and NGF mRNA in gallbladder adenocarcinoma were significantly higher than those in paraneoplastic tissues and chronic cholecystitis (P<0.01) .The positive rates and scores of NGF and NGF mRNA were significantly lower in the cases of adenomatous canceration or high-differentiated adencarcinoma, maximal diameter of mass≥2cm, metastasis of lymphnode, and invasion of regional tissues (P<0.05 or P<0.01) .Conclusion The expressive levels of NGF and/or NGF mRNA might be important biological markers for reflecting the carcinogenesis, progression, biological behaviors and prognosis of gallbladder adenocarcinoma.
