奥曲肽对肝星状细胞胶原合成及TIMP-1、MMP-2表达的影响

Objective To investigate the effect of octreotide on the expression of collagen、TIMP-1 and MMP-2 in cultured rat hepatic stellate cell (HSC) in vitro.Methods HSC cultured in vitro were randomly divided into 4 groups and different concentrations of octreotide were added to the cell groups.The synthesis of collagen type Ⅰ and type Ⅲ was measured by enzyme-linked immunoaddsordent assay (ELISA) , the expression of TIMP-1 and MMP-2 was detected by semi-quantitative RT-PCR.Results Octreotide remarkably reduced the production of collagen type Ⅰ and type Ⅲ.Octreotide down-regulated the expression of TIMP-1 mRNA, while up-regulated the expression of MMP-2 mRNA. (all P<0.05) .Conclusion Octreotide could significantly reduced the content of collagen type Ⅰ and typeⅢ.Octreotide can promot the expression of TIMP-1 mRNA, while inhibite the expression of MMP-2 mRNA.All of these biological effects of octreotide on HSC were associated with recovery from liver fibrosis.
